Abstract
İyi bilinen bir karsinojen olan 4-dimetilaminoazobenzen’in (DAB) embriyonik sıçan hücrelerinin mitokondriumları ve mitokondrial Isositrat (ICDH) ve glutamat (GLDH) dehidrogenazlar üzerindeki tesiri histokimyasal olarak araştırılmıştır. 0.1 mg DAB ihtiva eden vasatta embriyonik sıçan hücreleri 1.5, 3, 5 ve 24 saat inkübe edildiklerinde, yüksek enzim aktivitesi gösteren mitokondriumların sayılarının arttığı müşahade edilmiştir. Vasattan DAB nin çıkarılması halinde bilhassa ICHD aktivitesinin aşağı yukarı normal hücrelerde görülen seviyeye düştüğü görülmüştür, ki bu da hücrelerin ve enzimlerin ortamdan DAB nin kalkmasıyla yeniden eski hallerini alabildiklerini göstermektedir.
The effect of 4-dimethylaminoazobenzene (DAB), a well known carcinogen, on the mitochondria of the rat cells and on the mitochondrial dehydrogenases was investigated histochemically. Isocitrate (ICDH) and glutamate (GLDH) dehydrogenases were the selected mitochondrial enzymes for investigation. Incubation of embryonic rat cells in medium containing 0.1 mg of DAB for various hours; namely 1.5, 3, 5 and 24 hours; resulted in increased number of mitochondria exhibiting higher enzyme activity. Removal of the DAB from the medium resulted in decrease of the enzyme activity down to approximately normal level, especially in the case of ICHD; indicating a recovery of some sort.
